Ronaldo showed little mercy, however, stroking his spot-kick into the bottom corner for his 42nd goal of another prolific season.
Real were soon reeling as Ramos was sent for an early bath after the slightest of contact sent Neymar tumbling to the floor in the penalty box on 65, with Messi matching Ronaldo by guiding his spot-kick into the corner of the net.
Benzema was surprisingly withdrawn as Ancelotti looked to bolster his backline by sending on Raphael Varane, while Neymar made way for Pedro.
The game continued to ebb and flow but it was Barca who came closest to netting a seventh goal of the game, with Dani Alves thumping a low 18-yard strike against the upright when Lopez was beaten on 74 minutes.
Barca did make it 4-3 with five minutes remaining when Messi completed his hat-trick from the spot after Xabi Alonso clumsily brought down Iniesta.
Atletico Madrid's 2-0 win over Real Betis coupled with Barca's victory means the La Liga title race is building to a superb climax as Real and Atletico are locked on 70 points, with Barca now just one point behind in third place.
